New cancer Vaccine Makes Immunotherapy Effective for 75% of cancer patients

If today immunotherapy works only in 20% of cases, then with the help of a new vaccine, 75% of patients with tumors can be effectively treated. It is based on a drug that is approved for use in humans. Breakthrough therapy is already being tested in pilot clinical trials.

Article by Koerner et al. PLGA-particle vaccine carrying TLR3/RIG-I ligand Riboxxim synergizes with immune checkpoint blockade for effective anti-cancer immunotherapy published in the journal Nature Communications – VM.

In recent years, there has been a major breakthrough in oncology thanks to the development of an immunotherapy method that aims to suppress immune control points. Checkpoint inhibitors stop the decline of the immune response and instead support the attack on tumor cells. This therapy is very effective in melanoma, squamous cell skin cancer, lung, kidney and liver tumors, but it works only in a small number of patients.

German scientists from The University of Constance has been working on this problem and is now presenting a new experimental vaccine that can increase the effectiveness of immunotherapy by almost four times, according to a press release from Combining immunotherapies against cancer.

The vaccine is based on the immunostimulator riboxime, approved for use in humans. Riboxime causes a T-cell response necessary for the effectiveness of drugs that block immune checkpoints, scientists explain.

Testing of the combined approach on mouse models showed that the therapy increased its effectiveness by up to 75%. The experiments included models of prostate, breast, and melanoma cancers.

A strong antitumor immune response persisted even for eight weeks after treatment, and to achieve this effect, scientists used very low dosages of the vaccine.

Currently, the team has already launched a pilot phase of clinical trials. According to their estimates, the new vaccine in combination with immunotherapy will significantly improve the treatment of certain types of tumors.
