Chlamydia? Mycoplasmosis? Try gene therapy!

Antimicrobial peptides isolated from arthropod poisons are promising competitors to conventional antibiotics.

However, their introduction into the body is associated with a number of difficulties (for example, they can decay there ahead of time, they are expensive to synthesize, they can cause allergies). Therefore, Russian scientists have developed a new approach: genes encoding antimicrobial peptides are introduced into animal cells. When faced with an infection, these genes can be activated, and the cells themselves will begin to produce the drug. Vasily Lazarev, head of the Laboratory of Genetic Engineering of the Research Institute of Physico-Chemical Medicine, told more about the method at the conference "Postgenomic methods of analysis in biology, laboratory and clinical Medicine", held in Moscow on November 17-19, Informnauka reports.

Antimicrobial peptides are a component of the innate immunity systems of animals, plants and fungi. These substances bind to the membranes of bacteria and damage them. Scientists have long considered antimicrobial peptides as an alternative to antibiotics, but their use is associated with a number of difficulties, first of all, we are talking about the delivery of the active substance to the area of inflammation. Like all foreign substances of a protein nature, antimicrobial peptides are destroyed in the animal's body, and it is difficult to achieve their effective concentration in those areas where bacteria multiply.

The staff of the Research Institute of FCM and the Institute of Bioorganic Chemistry proposed to introduce into the body not the antimicrobial peptides themselves, but the genes in which their amino acid sequences are encoded. This approach has already shown good results in animals.

Scientists have worked with mycoplasma and chlamydia. These two kinds of bacteria are able to parasitize inside the cells of higher organisms, which makes treatment very difficult. But they are well susceptible to antimicrobial peptides, in this case – to mellitin from bee venom and latarcins from spider venom. Researchers have created a genetic vector containing an antimicrobial peptide gene and a system to regulate its production: the peptide is synthesized in cells not constantly, but only when the antibiotic doxycycline is added (this antibiotic itself does not act on chlamydia and mycoplasma).

After the system was tested on cell culture, the researchers began testing the method on animals. They worked with one type of chlamydia and two types of mycoplasma. The causative agent of avian mycoplasmosis (Mycoplasma gallisepticum) infected chickens. Two human parasites were tested on mice, for which it was necessary to artificially transfer bacteria from one mouse to another until natural selection led to the emergence of strains capable of effectively infecting an atypical host.

It turned out that timely activation of the production of antimicrobial peptides in animal cells leads to a complete cure of two thirds of them and to a significant reduction in the infectious load in the remaining third.

Vasily Lazarev does not exclude that in the future this technique can be used for the treatment of chlamydia and mycoplasmosis in humans, but emphasizes that the most important area of application of the new approach is agriculture. Mycoplasma gallisepticum causes epidemics of severe respiratory disease in birds and causes huge economic damage to poultry farming. Today, when the disease is detected, all birds on the farm are destroyed so that the disease does not spread to other farms. The introduction of a new technology will allow you to simply spray a suspension of genetic vectors in chicken coops. They will penetrate into the cells of the pulmonary epithelium of birds, and the disease will cease to be a serious threat: when it is detected in the first bird, the rest will simply add an antibiotic activator to the feed, antimicrobial peptides will begin to be synthesized in their cells, and the disease will be defeated.
