Scientists at the University of Alabama at Birmingham (USA) have managed to age mice and rejuvenate them by controlling the activity of genes in mitochondria. The scientists' article was published in the journal Cell Death & Disease (Singh et al., Reversing wrinkled skin and hair loss in mice by restoring mitochondrial function).

Researchers have introduced a mutation into the DNA of rodent mitochondria that affects the POLG gene encoding the enzyme DNA polymerase gamma. This compound is localized in the mitochondria and participates in the replication and repair of mitochondrial DNA (mtDNA). It has been shown that damage to POLG causes mitochondrial dysfunction, in particular, dysfunction of oxidative phosphorylation, which contributes to less efficient energy storage. In turn, this leads to the development of signs of aging, for example, the appearance of wrinkles and hair loss.

Scientists have added an additional copy of the POLG gene with a mutation to the fertilized egg cell of mice. This defective DNA begins to participate in the production of polymerase only in the presence of the antibiotic doxycycline. Doxycycline began to be added to food and water consumed by rodents when the genetically modified animals reached the age of eight weeks. After two weeks of taking the antibiotic, the mice began to show signs of aging. The black fur began to turn gray and fall out, the skin began to shrivel, and the spine began to bend.

Stopping the consumption of doxycycline, which blocked the activity of the mutant copy of POLG, increased the content of mitochondrial DNA. This contributed to the weakening and almost complete disappearance of the symptoms of aging in rodents.

According to scientists, the observation of the rejuvenation process in animals during the restoration of mitochondrial function was carried out for the first time.
